Take approximately 20 mL of urine sample and place it into a 50 mL plastic centrifuge tube. Centrifuge for about five minutes. Transfer 2 mL of the clarified urine sample into a 10 mL glass test tube with a stopper, and add 50 μL of 4000 mg/L heptadecanoic acid internal standard. Add 1 mL of 5% hydroxylamine hydrochloride solution, vortex mix, and allow the oximation reaction to proceed at room temperature for 60 minutes. After the reaction is complete, add an appropriate amount of hydrochloric acid to adjust the pH to 1, add approximately 1 g of sodium chloride, vortex to saturate the aqueous layer, and extract twice with 3 mL of ethyl acetate. After allowing the mixture to stand and separate into layers, combine the extract into the same glass test tube (accurately transfer a total of 5 mL of extract). Then, evaporate the solvent slowly at 50°C using the nitrogen evaporator. After evaporation, add 200 μL of derivatization reagent (BSTFA+TMCS 99:1), vortex mix, and place the mixture in an oven at 80°C for 30 minutes for the derivatization reaction. After the reaction is complete, transfer the solution to a 2 mL small sample vial, and then take 1 μL for GC-MS analysis.
